3 dimensional characterization of habenular depression molecular markers

Anatomically and functionally habenula links the forebrain with the midbrain structures that are involved in the release of monoamines and known to play a key role in the pathophysiology of depression. The habenula consists of heterogeneous cell populations and many attempts to compartmentalize the habenula according to the cell type have been performed using 2 dimensional tissue sections. However, for the investigation of the role of habenula in pathophysiology of depression, 3 dimensional characterization of habenular depression molecular markers is necessary. In this study, I set up 3D reconstruction methods for mice habenula, and performed volumetric analysis and molecular characterization of lipopolysaccharide (LPS) depression model habenula. As a results, I found that LPS depression model mice showed smaller habenula volume than control mice. Interestingly, this volume reduction was prominent in MHb(medial habenula) but not in LHb(lateral habenula). Furthermore, the expression of Tac1 (dMHb specific marker) was decreased in all parts of dMHb, ChAT (vMHb specific marker) was decreased in rostral and middle part of vMHb, and Tacr1 (Hb marker) was decreased in rostral and middle part of MHb and LHb. These results provide evidence that medial habenula plays important role in LPS depression model and three-dimensional analysis method for habenula (volumetry and gene expression) established in this study will be useful for further investigation of the role of habenula in pathophysiology of depression.
