Abstract It has been reported that copper and zinc are virulence factors in pathogenesis of Aspergillus fumigatus. Mac1 is well-known transcription factor in copper homeostasis of Saccharomyces cerevisiae and mrpA of A. fumigatus is a homologue of Mac1. We investigated the function of mrpA on zinc metabolism to figure out how mrpA associated with metal homeostasis. mrpA deletion strain exhibited growth defect on Aspergillus minimal medium (AMM) and complex medium (CM) both. Interestingly, zinc limitation suppressed growth defect of mrpA deletion strain. And expression of the genes encode zinc transporters significantly reduced compared with wild type when mrpA was deleted. In addition, mrpA deletion strain showed growth defect to the cell wall stress-inducing agents. Superoxide dismutase (SOD) activity was reduced when the mrpA was deleted and mrpA deletion strain was sensitive to H2O2. Taken these results together, we suggest that MrpA monitors the environmental change of zinc concentration as concerted manner with copper homeostasis in A. fumigatus.
