Fabrication Techniques and Testing of Microfluidic Biochips : Preconcentrated Electrospray Ionization Interface for Mass Spectrometry

Electrospray mass spectrometry has been shown to be a suitable method for the analysis of small amounts of proteins. This study has demonstrated the feasibility of the coupling between preconcentration microfluidic device and electrospray ionization for mass spectrometry. In sequential mode the device showed high quality electrospray at 3000V with a intensity ratio of x260 and in continuous mode the device showed a x 13 intensity ratio at 2500V and both modes showed high quality electrospray suitable for mass spectrometry applications. This work suggests an example of the concept of 'Lab on a chip' and presents the possibility of multifunctional microfluidic chips. Microfabrication technologies were used to construct devices which includes preconcentration components and an electrospray tip. The tip was mechanically cut followed by a mono-layer coating process of a hydrophobic substance to minimize the dead volume of the Taylor cone. The successful coupling online of micro fabricated devices to an electrospray ionization MS represents an essential step toward the construction of automated, high-throughput, and high sensitivity analytical systems.
