Overexpression of Thymosin beta prevents Motoneurons from Programmed cell death in the Chick embryo

The thymosin-beta (Tβ) is a small cytosolic polypeptide which acts as a regulator for actin dynamics and expresses transiently on embryonic central nervous system. However, it is little known about the physiological function of Tβ in the nervous system. To further assess the roles of Tβ in the nervous system development, YFP-tagged Tβ was introduced to the developing chick spinal cord using in ovoelectroporation. Using this method, I was able to successfully expressed Tβ on >30% of spinal motoneurons (MNs). Interestingly, there was a significant reduction of dying cells (pyknosis) in Tβ+ side during the programmed cell death period (E7) of MNs, compared to that in YFP+ control MNs. Moreover, pyknosis reduced and survival increased on spinal MNs of E6 after deprivation of target, and the phosphorylation of c-jun, a sign for early cascade of apoptosis pathway, was reduced at E7 on the Tβ transfected side. Mutant Tβ, which has mutant actin binding domain, also reduced pyknosis of MNs at E7. Taken together, these results suggest that Tβ15 may be involved in the regulation of developmental PCD of MNs, independent of trophic factor from targets.
