ICAM3 (Intercellular adhesion molecule-3) is the Ig-like cell adhesion molecule, mostly expresses on human leukocytes and involves in cell-cell interactions. The function as a potent signalling molecule of ICAM3 has not been well understood. Recent studies showed that ICAM3 is involved in maintaining cell survival. So, we examined that the cell proliferation was affected by increased expression of ICAM3 in H1299 lung cancer cell line. We have observed the phosphorylation of FAK, Akt, MAPK pathway through PTEN inhibiton by ICAM3-overexpression, resulting in subsequent leading to down-stream survival signal initiation. We examined the ICAM3-induced cell growth by BrdU incorporation, colony-forming assay, FACS analysis, and cell counting. In addition, we observed that ICAM3 enhanced tumor foci formation by transformation in NIH3T3 cell line, demonstrating that ICAM3 plays a role in tumor proliferation. From the results, we suggest that increased expression of ICAM3 might contributes to tumorigenesis and enhanced cell proliferation through activation of Akt signalling. Therefore, our concepts for ICMA3 could opens new possibilities for the diagnosis, prevention, and treatment of tumors.
