들깻잎(荏子葉) 열수 추출물이 간 보호에 미치는 효과 : Water extract of Perilla frutescens leaves exhibits hepatoprotective activity in vitro and in vivo

Perilla (Perilla frutescens (L.) Britt. var. japonica (Hassk.) Hara) is one of the most common herbs in Korea. These leaves are also used as medicinal plants in East Asian countries and for protection against food microbial poisoning. In this study, we describe its in vitro antioxidant activity. The water extract of perilla leaf possessed antioxidant activity, free radical scavenging properties assayed using 2,2-diphenyl-1-picrylhydrazyl (DPPH), and reducing activity using ferric reducing antioxidant power (FRAP) assay. The cytoprotective effects of Perilla leaf were demonstrated in water extracts of freshly isolated primary rat hepatocyte system. Tert-butylhydroperoxide (t-BHP) is a well known inducer of oxidative stress in biological systems. The cells cytotoxity studies by diphenyltetrazolium bromide (MTT) and lactate dehydrogenase(LDH) leakage and malondialdehyde(MDA). The cytochrome P450s and other enzymes were investigated following the administration of oxidative stress. The water extract of perilla leaf exhibited radical scavenging activity with an IC50 of 400.5±3.5㎍/㎖. The antioxidative activity of Perilla extract(62.5~1000㎍/㎖) against t-BHP(0.5mM for 2hr) showed a 5-fold decrease compared with t-BHP alone in LDH leakage. Furthermore, malondialdehyde formation was significantly decreased. The in vivo study showed that the pretreatment with PLE (1000 and 3000 mg/kg) by gavage for 5 days before a single dose of t-BHP (ip; 0.2 mmol/kg) significantly lowered serum levels of the hepatic enzyme markers (alanine aminotransferase and aspartate aminotransferase), and reduced oxidative stress of the liver, such as GSH content and lipid peroxidation in the liver in a dose-dependent manner. Histopathological examination of the rat livers showed that PLE reduced the incidence of liver lesions, including hepatocyte swelling, leukocyte infiltration, and necrosis confluent induced by t-BHP.
