초록/요약
The major problem in the therapeutic management of bladder cancer is tumor invasion, resulting in the tumor metastasis. Although the genes orchestrating the process need to be identified, involvement of a specific gene, osteopontin (OPN), in bladder tumor invasion has to be clearly elucidated in order to understand the mechanisms of tumor invasion. Here, we show that OPN-treated cells showed increased in vitro tumor invasion. The increase correlated with activation of mitogen activated protein kinase (MAPK) and Jun NH2-terminal kinase (JNK), and the in vitro tumor invasion was inhibited by treatment with inhibitors of these kinasees. Also, the amounts of pro-inflammatory cytokines secretion were increased in OPN-treated T24 cells. In real-time PCR analysis, the expression of MMP-2 and MMP-9 mRNAs was increased, and the increase was counterbalanced by treatment with 6-aminoquinazoline (NF-kB activation inhibitor II). This novel OPN-mediated tumor invasion via cell signaling pathway plays a critical role in tumor invasion of bladder cancer cells.
